In-vitro Reconstitutions to understand cellular functions

The bottom-up approach is based on the physicist Dr. Richardt Fynmann’s the philosophy of building is understanding. The cellular functions are recreated from the purified proteins to understand the entire functionality of the key components and have a comprehensive understanding. The interdisciplinary research is gaining interest among the scientific community specifically the ones interested in building a synthetic cell. The in-vivo experiments mostly involve knocking down or overexpressing the proteins and making observations of the changes that may result on a cellular function of interest. The influence of the cellular complexities on the observations in the in-vivo experiments cannot be ruled out. Often the functions of a knockdowned proteins can be taken over by the other proteins in the in-vivo environment, imposing an experimental limitation and making it difficult to give accurate interpretations. Therefore, the bottom-up approach gives an advantage over the in-vivo experiments to exemplify the hierarchical interactions involved among various components of the cellular function and give a comprehensive understanding. In my talk I will be presenting some of my own research work to show how the bottom-up approach can be employed to explain interactions at the molecular level and pattern formation at the cellular level.
