Abstract:. Axons, tubular protrusions of neurons, play a crucial role in various physiological phenomena. Its structural integrity and proper functioning are maintained by key biopolymers such as the membrane periodic skeleton (MPS), microtubules, and neurofilaments. These biopolymers can undergo structural changes following injury, which may affect axonal function. Therefore, it is crucial to understand their role in axonal mechanics during injury. To study this, we developed a custom laser ablation setup using a high-energy, picosecond UV pulsed laser with a tightly focused beam to mimic axonal injury. Axons were ablated at the midpoint using two approaches: full ablation, which severs the axon into two segments, and partial ablation, which disrupts internal components while preserving the membrane. Distinct retraction responses were observed: full ablation led to segment retraction and buckling, while partial ablation caused inward retraction of internal material. We analyzed these mechanical responses under various pharmacological conditions and also explored the role of calcium in axonal retraction. Furthermore, in calcium-chelated media, we investigated the recovery dynamics of both actin and microtubules during axonal resealing.
